Help with notes of the fretboard.

Im trying to learn the individual notes on the fretboard and have made myself a little chart to study from.
So the question i have is that if a tab that says to play like this.

the notes being played are d# and g (i hope im right)

Instead of playing that way could i play it like this, As its still the same notes.

That is right.
To controll if you're playing two identical notes you can just pick them both at the same time and you'll hear if it's right.
